A Colorimetric CO2 Hydration Assay for Facile, Accurate, and Precise Determination of Carbonic Anhydrase Activity

A simple, reliable, and universal method is demanded for routine determination of carbonic anhydrase (CA) activity, overcoming the limitations previous assays that are inaccurate, complicated, expensive, or limited to a specific enzyme family. The most widely used Wilbur–Anderson assay was modified improve speed, accuracy, precision by employing temperature controllable UV/Vis spectrophotometer pH indicator phenol red. experimental setting, measurement, data analysis were facile straightforward. validated using commercially available bovine CA, showing obtained activity directly proportional amount enzyme. measured (2540 WAU mg−1) agreed well with previously reported data. comparison results esterase showed CO2 hydration should not be substituted in measurement CA activity. simple reliable colorimetric can adopted substituting traditional assay.
